No.
John 10:29 says
"My Father, who has given them to me, is greater than all, and no one is able to snatch them out of the Father's hand."
Once you truly Trust in Christ as your Savior, you can not loose your salvation. Also, and leaning more toward your question, The Bible seems to imply in places that the Holy Spirit is always with the Christian.
Also, these two verses are noteworthy:
"13In him you also, when you heard the word of truth, the gospel of your salvation, and believed in him, were sealed with the promised Holy Spirit, 14who is the guarantee of our inheritance until we acquire possession of it, to the praise of his glory."
Ephesians 1:13-14

Yes!!
Although relatively few in this lifetime will be called to Christ by the Father (John 6:44), known as the 1st fruits who recieve the gift of the Spirit of God upon repentance and baptism (Acts 2:38), there still is FREE WILL. Now if an individual who has been called/saved does not remain strong to the end and falls backward, willfully sins and refuses to repent, then that individual will lose his one and only chance at Salvation. In effect, that person has rejected the one and only covering for sin, Jesus Christ - there is no other covering for sin! By doing so , he/she will stand before Him at the 3rd Judgement and be charged, found guilty with nothing to pay for these sins, and will be cast into the Lake of Fire with the other unrepentant, incorrigible sinners. Pauls speaks to this in Hebrews 10:26.
Receiving God's Holy Spirit (aka the Spirit of Christ) to 'begetting' us in walking the way Christ walked (our example) is not a 'get out of jail card' to be abused. We must seriously change from our old, sinful ways and move forward in the faith of Christ doing as He did in all things. We will fall short but are promised forgiveness upon repenting. Continue to sin and refuse to repent and lose the gift of God's Spirit!!
