That depends on the temp of your water to start with, but at minimum, 30 min after it's been placed in the fridge.
You can check it after it's been in the fridge for 30 minutes, and if you want it firmer, check it every 10 to 15 minutes until it's as firm as you need.

You make up the mixture for the first colour that you want the jelly to be, and pour a layer of it into the required container. You leave this to set completely, then make up the colour of jelly that you want the next layer to be. Pour a layer of this jelly on top of the first layer of jelly that has now set.
