"Do you speak Spanish" would come in two forms, depending on to whom you are speaking. For a formal tone, one would use "¿Habla usted español?". Informally, it would be "¿Hablas español?"

There are several ways to ask if one speaks Spanish. If you are asking a friend or someone younger, you would say "¿Hablas español?" If you are asking an elder, family member, teacher, etc. (someone of superiority), you would say "¿Habla español?" If you are asking multiple people of which they are all friends or younger, you would say "¿Habláis español?" Finally, if you were asking multiple people in which at least one is of superiority, you would say "¿Hablan español?". But the latter can be formal and familiar plural in Spanish America, but only formal in Spain, since the personal pronoun "vosotros, as" is not currently used in The New World.
