To drag a file into iTunes first make sure that the file is in a format that iTunes will recognise otherwise it will not accept the dragged file.

You highlight the song in the main library, not a playlist, and then press delete. A pop-up window will appear, and instead of pressing keep file, say move it to trash, or delete.
