DNA in prokaryotic cells is usually circular while DNA in eukaryotes is usually linear. DNA in prokaryotes usually has fewer portions that dont code for something while eukaryotes usually have quite a bit of DNA sections that dont code for anything. The number of actual base pairs (the units that makeup DNA) that makeup eukaryotes is usually quite a bit more than the number in prokaryotic DNA.

In a prokaryote cell the DNA is located just within the cell, floating in the cytoplasm, whereas the DNA in eukaryote cells is located inside the nucleus.

Prokayotes usually have circular DNA, which is found in an area of the cytoplasm called the nucleoid. This, unlike the nucleus of eukaryotes, is not enclosed by a membrane.

Eukaryotes have linear DNA, which is contained within the nucleus of the cell.

In prokaryotes the DNA is in a circular chromosome free of all introns but still double stranded. Also there can be circular and small plasmids acquired by conjugation with quite a few less genes.

Eukaryotes have their DNA arranged in chromosomes, Structures wrapped around histone proteins and packed tightly for replication, or loosely as they normally are and then called chromatin. The chromosomes are sometimes methylated for inhibiting of gene purposes and genes are strung along the chromosome like beads. Many intronic regions separate the sense of the genes on the chromosomes, the exon regions.

What makes a eukaryotes different from a prokaryote?

There are about eleven differences between a prokaryote and eukaryote. The most basic difference is that the eukaryote have "true" nuclei containing their DNA, but genetic areas in a prokaryote isn't membrane-bound. If you need more differences between a prokaryote and eukaryote, ask me on my message-board.
