It takes 2.5 to 4 hours, it all depends on the traffic and time of day. In general it's better to use the Lincoln or Holland tunnels to cross into New Jersey as compared to traveling via Long Island and Staten Island. If getting out of Manhattan, and through the tunnels in the 15-20 minute range, you can expect to be in Atlantic City in 2 hours after that. Best route: turnpike to exit 11, then parkways to exit 38, then easet on the AC expressway until you see the brights lights and smell the ocean air. :)

Depending upon one's starting point in Ocean City and ending point in Atlantic City the drive from one island to the other is about 15 minutes. Longer if you are starting further south and heading further north.
