They both APPEARED (not necessarily starred) in the following films:
Two Fisted Law
Texas Cyclone
Baby Face
Dakota
Red River
Rio Bravo
How the West Was Won
They both appear in Rio Bravo and The Sons of Katie Elder. True John Wayne westerns:
In Rio Bravo, they are lawmen trying to keep a prisoner, who is awaiting trial for murder, in custody while the bad guys brother is trying to break him out and not letting anyone else get in the town to help. Dean Martin plays a classic drunk deputy who learns how to be sober. John Wayne is the sheriff trying to keep everyone safe until the judge gets there. Ricky Nelson (the singer) is also in this movie who becomes involved in this as a deputy and is a good shot. And a young Angie Dickinson is Wayne's love interest.
The Sons of Katie Elder has both John and Dean as 2 of 4 brothers with Earl Holiman and Michael Anderson Jr. as the other 2 brothers. They all get together for their mother's funeral and to settle her estate. They have all gone their separate ways and John is a hired gun. John is the one who realizes that the land that their parents owned was swindled from them and he (and the others) want to set things straight plus have money to make the youngest son Bud (Michael Anderson Jr.) go back to college. The bad guys who own the land and a lot of the town, frame the boys for stealing and are going to see them killed but the boys have other ideas. Unfortunately one of the brothers is killed, Bud is wounded badly, Dean is shot in the back and John saves the day in the end. But you figure Bud and Dean do survive in the end and John feels that he did right by their mother.
They are both really good westerns and worth seeing more than once.
They appeared in eight movies together... Rio Bravo, Red River, How the West Was Won, Dakota, Three Godfathers, Baby Face, Texas Cyclone, and Two Fisted Law
I think that they were friends, and had a mutual respect for each other.

John Wayne never co-starred with anyone named Diane... the closest I can find is Diana Muldaur (McQ) and Diana Gibson (Adventure's End)

I'm going to presume your question is, "Did anyone who was later a member of the Three Stooges ever appear in a western with John Wayne?"Shemp Howard appeared in Pittsburgh , which starred Wayne but was not a western.Other than that, no actor from the Three Stooges was ever in a film with Wayne.
Wayne Rogers starred in MASH for three seasons.
Later in his career Wayne made a few guest appearances on popular TV shows such as Rowan and Martin's Laugh-In, I Love Lucy (Lucy and Ethel stole Wayne's footprints from Grauman' Chinese Theater), and Maude (does anyone else remember the 'hairy coaster'?). He also made a public announcement commercial on the importance of regular cancer screening and early detection. But Wayne never starred in a TV series. It's possible that you may be thinking about a couple of Wayne's friends. Ward Bond who was friends with Wayne since their college days at USC starred on The Wagon Train until his death in the late 1960s. Or James Arness, who many thought resembled Wayne, starred on the long running series Gunsmoke. He did record an introduction for the "Gunsmoke" series. It played prior to the first episode, introducing James Arness to the television audience.
