Answer 1: Most theologians agree that this passage in Luke speaks of Mary, Jesus' mother and Jesus's half-brother James (James father was Joseph apparently and God was Jesus'). Mark 6:3 says, "Is not this the carpenter (Jesus), the son of Mary, and brother of James and Joses and Judas and Simon? Are not His sisters here with us?" And they took offense at Him. So, Mary had children after Jesus. The Greek word could mean "cousin" but putting Scripture together, Jude (Judas in Mark 6:3) was the half-brother of Jesus who wrote the small book of Jude and James wrote the very early New Testament book James.

Answer 2: Mark and Luke depict a "Mary mother of James" (Mark 16:1, Luke 24:10) after the crucifixion, who's described by Matthew as "the other Mary" (Matt 27:61). In earlier passages, Matthew and Mark had mentioned that she was also the mother of a "Joseph/Joses" (Matt 27:56, Mark 15:40 & 47). Despite the fact that the first "brothers" of Jesus named in Mark 6:3 and Matt 13:55 were "James (the Less) and Joseph/Joses", scholars (even those who don't assent to the 'cousin theory' of Jesus' brethren) agree that this passage does not refer to Jesus' mother because, after Jesus' death, Luke still describes Jesus' mother as "Mary the mother of Jesus" (Acts 1:14) and because such an oblique reference would be "unparalleled" in the NT, according to John P. Meier. Instead, Meier takes the same order of the names "James and Joseph" in Matt 13:55 and 27:56 and Mark's atypical use of "Joses" (compare Mark 6:3 & 15:40 & 15:47 with Mark 15:43 in the KJV) to be nothing more than 'striking' coincidences, while the likes of Joseph Blinzler instead take them as proof of the 'cousin theory'. In reference to the closing paragraph of the above answer, the opening verse of the Epistle of Jude reads: "Jude, a servant of Jesus Christ and a brother of James". This is also used as evidence for the 'cousin theory', since he doesn't describe himself as a brother of Jesus. The question of the genealogy of Jesus' brethren remains unresolved, though.
