According to Lauren Faust Scootaloo is not related to Rainbow Dash in any way, she just admires her a lot and is willing to do anything she says. They are viewed by the fanbase, and it has been vaguely shown in the show that Dash acts as Scotaloo's surrogate sister, however.

∙ 12y agoYes, in G3 Scootaloo and Cheerilee are sisters. However, in G4 (friendship is Magic) they probably couldn't be considering that Cheerilee is an earth pony while Scootaloo is a pegasus. Also, Cheerilee is Scootaloo's teacher and hasn't shown any relationship with her beyond that of a normal teacher.
