Short answer: No.
The bus station is located in the Port Authority in Midtown Manhattan, perhaps the most crowded area of NYC on any given day. Parking on the street is fairly impossible with the daily parking rules and lack of space. Even if you come across a parking garage with space for another car, it will cost you dearly.
I personally never drive into Manhattan unless it is a Friday or Saturday night (after the meters have expired and the weekend warriors are already home from work), and even then it will takes me a while find parking. If your tour is during the day, you may also want to factor in the ridiculous amount of traffic you will have to endure. Sometimes, the convenience of driving is more trouble than it is worth.
Your best bet is to park in Jersey and take a bus into the Port Authority or take the shuttle train. You may also find parking in lower Manhattan or in a less populated boro and take the train into Manhattan, but you MUST mind the parking signs, as this city wastes no time towing.
