The population of the Jamestown settlement varied in the 1600's due to several factors, specifically disease, starvation, and poor relations with native Americans. Originally 107 colonists came over to settle the colony. More colonists came over, bringing the total up to 500... but due to poor location and other factors (swampland that had stagnant water, which led to poor health) only 61 of the original settlers survived the winter of 1609-1610, known as "the starving time".
After much needed supplies and more settlers arrived from Lord De La Warr (Delaware later named after him in honor of saving Jamestown), the population stabilized and steadily grew. But in 1622, when the population was about 1,000, a surprise attack by the Powhatan tribe slaughtered around a third of the population, and again in 1644.
When the men arrived in 1607 there were 104, but in six months there were 34. The population stayed low due to disease, bad water, and starvation. In fact 1609 is referred to as the starving time because they were eating rats, leather, and each other.
What became known as the "Starving Time" in the Virginia Colony occurred during the winter of 1609-10, when only 60 of 500 English colonists survived

No one. Jamestown was founded in 1607 and was reorganized by John Smith in 1608. Well, less reorganized than increased the discipline of the colonists so that they'd actually work to keep themselves from failing.
