Your question is a good one as it involves several answers. I would say that it you want a reference because not only do you want to relocate, but you might be leaving the job, you might tell your current employer so that he or she can supply you with a good reference.
We could go on and on with this kind of question, but always remember you may want something from this employer one day, and if you were kind enough to tell them that you want to re-locate, you never know how they can help you in the future.
