Ang ekspedisyon ni Ruy Lopez de Villalobos ay isang misyon ng Espanya noong 1542 upang sakupin ang mga teritoryo sa Pilipinas. Si Villalobos ang unang Espanyol na naitalang nakapunta sa Leyte at Samar, ngunit hindi niya ito nagawang maangkin.
