Cities can be considered "major" for different reasons. Sometimes major means it has a very large population, other times it means it has the capital building, other times it means the presence of a university, and other times it may be a strategically important location.

So I will give you a few different answers:

New Orleans, Baton Rouge, Lafayette, Shreveport, Lake Charles, Alexandria, Houma, Thibodaux, Slidell, Mandeville, Covington

Baton Rouge (LSU, Southern), New Orleans (Tulane, Loyola, UNO, Xavier), Hammond (Southeastern LA U), Lafayette (ULL), Lake Charles (McNeese St), Grambling (Grambling St), Shreveport (Centenary College), Natchitoches (Northwestern St), Ruston (LA Tech), Monroe (ULM), Pineville (Louisiana College), and so on.

Along the coast there are many towns that are important for their role in fishing and petroleum and farming.

For exampe: Morgan City - Petroleum Industry, Shrimping Industry, Franklin - Sugarcane farming, and countless others.
