It can happen on the first dose; it can happen on the last dose, or anywhere in between. If you experience itchiness and a rash, swelling of the face/tongue/lips, stop taking it immediately, contact your doctor and take Benadryl. If you are swelling enough that you are unable to breathe, your throat is closing in (or wheezing like Asthma), or you feel faint - get to the ER/call an ambulance.

I just got one 4 days ago and it still hurts I'm not sure it's normal though, I can't sit on my left side or walk too hard on my left foot or elsde it hurts way too bad. There's a small bruise around where the shot was and it's red all around the size of my hand, AND it's got a bump around it the size of my hand. My boyfriend got the shot 3 days ago and it only hurt him for a day. Maybe I'm alergic to penicillin???

The effects of penicillin itself are not actually felt. The effect is that some bacterial infection will be cleared up and you will feel better instead of sick. The time for this to take effect could be hours or days depending on the dose of penicillin, the type of infection, the level of infection and the bacteria involved. These vary widely.

If it's real penicillin (Pen Vee), you can take it with food, so there is no waiting time before eating. If it's Cloxacillin or another penicillin that needs to be taken on an empty stomach, you should wait at least one hour.

One of the major side effects of penicillin is a rare but sever intestinal condition. This can occur days or even weeks after the penicillin injection.

You shouldn't be contagious after 24 hours of being on the antibiotic.

Penicillin has a short length of time within the system, so waiting a day would probably be long enough for it to have its effect.
