Causes for Lou Gehrig's disease are still a matter of research and considered largely unknown, although in at least 10% of cases there seems to be a significant genetic factor associated with mutations in various genes causal or related to production of one or more faulty enzymes. Glutamate is also known to have a role; protein misfolding seems to be significant, as do brain lesions which may cause the effects to propagate; some studies show promise in research of trauma and dietary issues.
