The saying "two's company, three's a crowd" refers to the phenomenon which can happen when a third person joins a group but doesn't really mesh with one of the other two. Alternatively, the saying could be in reference to the way people consider groups of people to be classified. To put it simplistically, a person views two people as an acceptable group, but three people is too many. This is similar to how some people consider something to be noise while others say it's music.
"ones and twos" refers to the two turntables commonly used by DJs. "Spinning on the ones and twos" then, is the act of playing (spinning) records on the turntables.
