It is a procedure to stain the bacterial capsule to study its presence and also morphology for this the following steps are performed:

Place a drop of Congo red on one end of the slide

Aseptically pick up a sample from a culture with your loop and mix briefly with the Congo red stain.

Spread the mixture across the slide

Air dry ONLY. Proceed to inoculate your tubes while drying.

Stain 1-2 minutes with the A-M Capsule Stain Solution. S(THIS STAIN CONTAINS PHENOL)!

Rinse very gently, air dry,

Look for red cells surrounded by a clear, colorless zone on a blue background.

1. Place a few drops of Congo red on a clean slide. Do NOT use a water drop in this sample preparation.

2. Mix in a small amount of culture.

3. AIR DRY. DO NOT HEAT FIX. (Heat fixing destroys protein capsules; it could also dehydrate the cells which would result in shrinking of the capsule)

4. Place the slide on a rack of the staining tray. Gently flood the smear with the Maneval solution and wait 5 minutes.

5. Lift the slide and gently pour off excess stain.

Rinsing with water is the next step. Rinsing must be done with great care. As the sample was not heat fixed to the slide, the sample preparation is fragile. Suggestions for careful rinsing:

· Place slide on bottom of staining tray.

· Add water to tray in the opposite corner from the placement of the slide. Add the water SLOWLY

until a large puddle forms.

· Tilt staining tray and allow water to rinse the slide. Remove slide from tray.

6. Place slide on absorbent paper. Allow sample to AIR DRY. Do not blot.

7. Examine the slide with an oil immersion lens at 1,000X magnification.

Since the capsule is water- soulube, copper sulfate, rather than water, is used to wash the purple primary stain out of the capsular material without removing the stain that is bound to the cell wall.

When a stain, such as an acid dye, cannot penetrate the outer layers of a microbe, the cell will appear transparent on a colored background. This stain is called a negative or background stain. It is performed by mixing the dye with a suspension of bacteria on a slide and spreading the mixture into a thin layer for viewing. The capsule is a structure surrounding the cell wall that certain bacteria can produce. The ability to form a capsule is genetically and environmentally controlled. The capsule appears as a colorless layer between the bacterium and the background.
