According to the super computer Deep Thought, the answer is "42". The problem was that no one knew what the question was. According to the book, another computer was created to discover the question, that computer being the planet Earth.
In the sequel, The Restaurant at the End of the Universe, it is discovered that the ultimate question of life, the universe, and everything (possibly slightly distorted) is: "What do you get if you multiply six by nine?"
While 6 x 9 = 54 in base 10, it does equal 42 in base 13, which author Adams claimed was completely serendipitous.
Previously, the designers of the computers had given up, and settled for the question "How many roads must a man walk down?" (from Bob Dylan's Blowin in the Wind)
