If you can, come up with a good answer fast.

If you can't think of a good answer, be honest. If it's a question about the company or yourself, say, "To be honest with you, I'm not sure how to answer that. I'd like to acquaint myself more fully with that topic (if it's a question related to the job or company) so that I could offer a knowleadgable answer."

Companies don't expect you to be perfect. The above response indicates a thoughtful person willing to do the reasearch necessary to reach a goal.
