There is not a great deal of difference. The verb 'answer' can be transitive as well as intransitive, whereas 'respond' and 'reply' can only be intransitive. 'Respond' can refer to contexts other than verbal, whereas 'answer' and 'reply' do not normally do so. In a written context, 'respond' may be seen as politer than 'answer' or 'reply' ('please respond within the next two weeks' sounds slightly less peremptory than 'please reply within the next two weeks', which in turn is less peremptory than 'please answer within the next two weeks').

For practical purposes, in everyday conversation, the words are fairly interchangeable.
