The leading strand is created continuously, but the lagging strand is created as small fragments, known as Okazaki fragments. These fragments are later joined together to form one complete strand.

The Leading strand is synthesized in the same direction as the movement of the replication fork, and the lagging strand is synthesized in the opposite direction. Also the Leading strand is synthesized continuously, whereas the lagging strand is synthesized in short fragments that are ultimately stitched together (okazaki fragments.)

The two strands of Double stranded DNA are in a Three-prime to Five-prime end-over-end head-to toe configuration. DNA may be synthesised in only the 5' to 3' direction - this is the Leading Strand; synthesis of the 'oppositely configured' Lagging strand occurs using 'Okazaki fragments'.
