What is the leading female singer in a opera called?

In the modern era (today) simply "female lead". in 18th and 19th centuries she would be callled "Prima Donna" (First Lady) taking this a step further a "Prima Donna assoluta" would be ahead of all the rest of the female singers. The only ones to be accorded that title in modern times are Maria Calla and Dame Joan Sutherland.
