Imagine that you have a test product and a reference product and Group 1 subjects are taking the test product and Group 2 subjects are taking the reference product. A two-way crossover would be Group 1 subjects being dosed with the reference product and Group 2 subjects being dosed with the test product. In essence, you have 2 groups of subjects that switch or crossover from the product they were taking to the product the other group of subjects was taking. Usually there's a time like a rest phase between the crossover to diminish the chances of carryover from the previous product. The advantages of this type of study is there is a better comparison of the test and reference products without having to be as concerned about variation between subjects. The results for Subject A taking the test product will be compared to the results of the same subject (Subject A) taking the reference product.
A one way cross over study is basically a trial design that is used in cases where two treatments are administered successively in the same group of trial subjects. While in two fold crossover studies subjects may start with any of the two treatments depending on which treatment is named first on the randomization list.
