Why is it advisable to start first with low power lens when viewing a slide?

It is easier to start with low power lenses because it makes it easier to find the object that you are focusing on and object looks clear . It is not easy to focus with high magnification .

How should you first focus on a specimen when using the microscope?

start by focusing with the lowest lens to make sure the slide is right and you can see the specimen them proceed to the next lens power and focus again just to make the view clear then proceed to the highest and focus one more time

Why is slide centered before moving to higher power on microscope?

When you switch to high power, you are focusing on a very small part of the large picture. If you center your object under low power (preferably by the little black arrow most microscopes have) you will see the object you are looking at 100% of the time. If you do not center your object, the object will end up being off to the side of what you are looking at in your field of view. It's kind of like zooming in on a map on the internet, you have to center what you want to zoom in on or else you will zoom in on an area you did not want to look at.

When do you use fine focus and the coarse focus knobs?

The fine focus knob is used to bring an object into fine focus, while the course focus knob is used to bring an object into approximate focus. The course focus knob should only be used on the low-power and scanning objectives of the light microscope. Using it on the high power microscope is okay, but you should be very careful. It is no recommended to use the course adjustment knob on the oil-emersion objective as it is so close to the stage and could lead to a potential break in the slide. Fine focus adjustment knob is useful to bring out the fine details in the specimen on your slide. Can be used on any of the objectives, but most receptive on the high-power and oil-emersion objectives.
