Take the Uptown A or C trains (of the A-C-E, the blue line) from 34th Street-Penn Station to 59th Street-Columbus Circle. Depending on which exit you use, you will get off the subway at West 57th to West 60th Street.

You could take the Uptown E train (of the A-C-E, the blue line) from 34th Street-Penn Station to Lexington Avenue-53rd Street. This station is connected to the 51st Street Station on the 6 local train (of the 4-5-6, the dark green line). Transfer to the Uptown 6, 4 stops, to 86th Street (at Lexington Avenue).

This way, you only have to make 1 transfer, but there is the inconvenience of having to walk 2 blocks underground to transfer.

Or, you could take the Uptown 1-2-3 (the red line), 1 stop, from 34th Street-Penn Station to 42nd Street-Times Square. Transfer at Times Square to the S shuttle train (color grey), east across 42nd Street, from Times Square (West 42nd) to Grand Central (East 42nd). Then transfer at Grand Central Station to the Uptown 4 or 5 express trains, 2 stops, from 42nd Street-Grand Central to 86th Street (at Lexington).

This route has the inconvenience of having to make 2 transfers, however, you don't have to walk underground between stations, plus, at Grand Central you can get the express trains (the 4 or 5).

So each option is pretty much equally good. It just depends on whether you'd rather make 2 transfers, or schlep 2 blocks from one station to another. I guess I would probably pick the first option (the E to the 6) if I were traveling at night (after around 11 PM), especially on a Saturday or Sunday night, because the trains run less frequently, so it's better to only have to wait for 2 trains, rather than 3. Plus, the express trains run local at night, so you wouldn't get the benefit of express service from Grand Central to 86th.

Otherwise it doesn't really matter.
