Two of his sisters died when they were babies. One died when she was eight. His sister Joan lived to the age of 77 and outlived Shakespeare. Nobody knows what the cause of any of their deaths was.

He actually had four sisters, two of whom died as babies. His sister Anne died at the age of 8, and his sister Joan lived to the ripe old age of 77. Joan probably died of old age; we don't know what the others died of.

William Shakespeare was John's third child. His first two were daughters who died as infants. The first died weeks after her birth which suggests complications arising out of the birth. The second lasted a bit longer, but would appear to have died from the plague.
