From the data estimated for total population in 1776, departing/loss of slaves during the revolution (1776-1783), the increased slave transport statistics of the 1780s, slave birth rates, and most importantly, the census of 1790, a reasonable "guesstimate" can be made. With an estimated population of 2.5M in 1776, ~380-400k were slaves (of all types to include Indian, but not including indentured servants).

There seems to be no clear estimate on the number of slaves in the American colonies in 1776.
