What would cause a large knot where the neck meets the spine on your back if you're a 29-year-old female and this developed almost over night?

It is very important that you visit a Chiropractor. Most likely your neck and spine are out of adjustment, or subluxated. Meaning, they aren't curving the way they should so it pinches a nerve which cuts off the "power supply" to areas of your body. It is like pinching a hose, the water can't come out properly. Your knot could be something more serious. Chiropractors manipulate the spine to correct this and many other problems.
My daughter had something very similar and after spending a lot of money at the chiropractor with little or no improvement, she is now seeing a physical therapist. She is double jointed and the ligaments were not supporting her neck so her neck muscle had to work overtime. She has a huge stress knot in the back of her neck and has been suffering with this for a long time. My suggestion is to see a physical therapist.
