William Henry Harrison (1773-1841) was only President for 32 days, and was the first US President to die while in office. Having contracted a viral disease, he died of pneumonia on April 4, 1841. His Vice-President, John Tyler, took the oath of office as the 10th US President on April 6.

William Henry Harrison gave such a long inaugural speech in the cold DC winter that he caught a cold which developed into Pneumonia. He died after serving less than a month as president, the shortest time served as president.

If you mean while in office than it would be William Henry Harrison who just one month before he died in office in 1841.

If you mean in his entire life , it was John Kennedy who was 46 when he was killed. Polk was the youngest to die of natural causes, at age 53.
