In terms of the actual fighting, the first shot fired was the so-called "shot heard 'round the world" which was fired by an unknown person (could be British or American) in Lexington, Massachusetts in 1775.

In terms of the grievances, both the Americans and the British faulted the other for a long list of historical issues, so it is unclear who started the American Revolution in that sense.
