Once the first sperm penetrates the egg, a series of changes take place in the protective layers around the outside of the main part of the egg (oocyte). This process, called the cortical reaction, makes the layers hard and not able to be penetrated by any other sperm.

When the sperm's plasma membrane fuses with the egg's plasma membrane, there is a depolarisation of the egg's plasma membrane caused by an influx of sodium ions into the cell. This acts as a temperary barrier (several minutes) to other sperm until the outer vitiline layer of the egg becomes solid!!

It's important only one sperm fertilises the egg because when one sperm penetrates the eggs, it causes the egg to build a wall to make sure only one sperm fertilises the egg. If more than one sperm penetrates the egg your egg cells divide and you may give birth to twins

You are asking about how pregnancy occurs, and it's quite an amazing process. During sexual intercourse, if there is no condom or Birth Control in use, the man's sperm enters the woman's vaginal canal. The sperm then swim through the vaginal canal and into the cervix. But while there are many sperm, there will only be one fertile egg and it will be in one of the woman's fallopian tubes, where it waits for 24 hours for one of the sperm to fertilize it. The sperm will all try to reach that egg, and it's almost like a competition-- with one winner, and all the other sperm are losers. When that one lucky sperm manages to reach and penetrate the fertile egg, that completes the fertilization process. Part of the process is that once the one sperm is successful, the other sperm are sort of locked out-- changes in the egg occur almost instantly, and no other sperm are able to enter.

The why of any question really can't be answered, it just is. That said, there are the same number of chromosones in a sperm cell and the egg. If more than one sperm got in, it could cause a miss match. DNA is like a zipper and the teeth have to match for it to work.
