So far, only four have been successfully assassinated. So let's go down the list:
1. Abraham Lincoln - he was shot by John Wilkes Booth, who was strongly pro-Confederacy and pro-slavery during the Civil War. Lincoln was President of the Union, and anti-slavery.
2. James Garfield - he was shot by Charles Guiteau. Guiteau was probably insane. He once wrote a speech in favor of Garfield during an election, and believed that the speech single-handedly won the election for him. When Garfield wouldn't give Guiteau a job in his Cabinet, Guiteau decided that God wanted him to murder Garfield.
3. William McKinley - he was shot by Leon Czolgosz, who was an anarchist- he basically wanted to overthrow the government and set up a country vaguely similar to communism as described by Karl Marx (not the communism that the Soviet Union had). There was a wave of anarchist assassinations of leaders across Europe at that time, and Czolgosz was inspired by this.
4. John F. Kennedy - this one's a little tricky. The official story is that he was shot by Lee Harvey Oswald, a former US Marine who was apparently a Communist sympathizer who had lived in Soviet Russia for a few years in his early 20s. However... I'm not going to get too much into it here, but there are people who don't believe the official story and think that there's a wider conspiracy that killed him.

Abraham Lincoln (16th President) was shot on April 14, 1865 in Ford's Theatre in Washington, DC, and died on April 15. His assassin, John Wilkes Booth, was a Confederate sympathizer who acted with the help of other agents from the South. Booth was shot on April 26 while being captured in Port Royal, Virginia.
James A. Garfield (20th President) was shot on July 2, 1881 in a Washington train station and died 11 weeks later on September 19. The man who shot him, Charles Guiteau, was mentally ill and had been denied a federal job in Garfield's administration. Guiteau was tried for murder, convicted, and hanged on June 30, 1882.
William McKinley (25th President) was shot on September 6, 1901 at the Pan American Exposition in Buffalo, NY, and died on September 14. The assassin, Leon Czolgosz, was an admitted anarchist who refused to openly testify at his murder trial. He was convicted on September 24 and died by electrocution on October 29, at Auburn Prison in Auburn, NY.
John F. Kennedy (35th President) was shot while driving in a motorcade near the Texas Schoolbook Depository in Dallas, TX, on November 22, 1963. His alleged killer, Lee Harvey Oswald, fatally shot a policeman before being captured hours later in a theater. Oswald, although claiming to be a "patsy," was a former Marine sniper and was known to have been angry about Kennedy's actions and inactions regarding Cuba. Oswald was shot and killed by Jack Ruby, a Dallas nightclub owner, while being moved to the county prison on November 24. His killing appeared live on national television.
