Yes. Though it seems it will remember it will keep an original backup (from the update from Windows XP SP1 to SP2 for example), but will erase other incremental updates since then. Do this only as a last resort if the virus that is being removed keeps coming back (likely from System Restore). Once you re-enable, create a manual restore point (from the System Backup utility)
